    Immunoglobulin VH Chain Gene Analysis of Peripheral Blood IgM-Producing B Cells in Patients with Kawasaki Disease

    PURPOSE: Kawasaki disease is a systemic vasculitis, and its etiology and pathogenesis are still not clear. Our study was undertaken to investigate the characteristics of the activation of B cells in the peripheral blood of Kawasaki disease (KD) patients and evidence of stimulation by superantigens. MATERIALS AND METHODS: Blood samples were obtained from three patients (2 males, 1 female) with KD, who were admitted to our Hospital, Seoul, Korea. The mean age was 1.2 years. Distribution of B cells was studied in the acute and subacute phases of KD patients. From the RNA of B cells, we obtained complementary DNA (cDNA) and performed polymerase chain reaction (PCR). To determine the oligoclonal expansion of immunoglobulin M (IgM) V(H) family, we cloned and sequenced the PCR products from each group and analyzed DNA. RESULTS: In the peripheral blood of acute phase patients, T cells were significantly decreased (p < 0.05), whereas B cells were significantly increased (p < 0.05). When the first PCR was done on the B cell chains, V(H)1 to V(H)6 were all found to be expressed. The number of micro gene clones obtained from 3 patients was 312, and they belonged to V(H)3, V(H)4 and V(H)5 family. M99686 germ line was most frequently used and the next most frequently used, were X92224/J, L21967 and L21964. A similar order was seen in patients. Among the clones, 20 sets of clones showed the same base sequence and this was frequent between V(H)2 and V(H)5. There was one set, which showed almost the same base sequence between different patients, and the homology was 99.5%. Twenty sets of clones that had the same base sequence showed high similarity to the germ line (94 - 100%). Among these, the clones that utilized the M99686 germ line were 4 sets which were most frequent. The 3-dimensional structure of one of these clones showed typical beta, sheet structure of immunoglobulin chains. CONCLUSION: The IgM transcripts expressed by the B cells in the peripheral blood of KD patients in the acute phase of the disease clearly showed an oligoclonal expansion, suggesting that KD is caused not by stimulation of a superantigen, but rather by a conventional antigen.ope

    Site Response Analysis Using Forced-Vibration Tests on Hydraulic-Filled Soil Deposit

    Forced vibration tests and detailed site characterization were performed at a geotechnical experimental site in Yong-jong Island where Inchon International Airport being constructed. The vibratory motions generated by the combination of hydraulic hammer compaction and dynamic compaction were monitored by 3-component velocity transducers in the down hole array as well as on the ground surface. Spectral analyses of the recorded vibratory motions were performed to evaluate the resonant frequencies of the experimental site. The linear analysis and simplified method were also performed to evaluate the resonant frequencies of the experimental site based on the results of detailed site investigation. The resonant frequencies of the experimental site evaluated by spectral analyses based on forced vibration tests were in good agreement with those of the linear analysis and the simplified method

    Transient receptor potential canonical type 3 channels control the vascular contractility of mouse mesenteric arteries

    Transient receptor potential canonical type 3 (TRPC3) channels are non-selective cation channels and regulate intracellular Ca2+ concentration. We examined the role of TRPC3 channels in agonist-, membrane depolarization (high K+)-, and mechanical (pressure)-induced vasoconstriction and vasorelaxation in mouse mesenteric arteries. Vasoconstriction and vasorelaxation of endothelial cells intact mesenteric arteries were measured in TRPC3 wild-type (WT) and knockout (KO) mice. Calcium concentration ([Ca2+]) was measured in isolated arteries from TRPC3 WT and KO mice as well as in the mouse endothelial cell line bEnd.3. Nitric oxide (NO) production and nitrate/nitrite concentrations were also measured in TRPC3 WT and KO mice. Phenylephrine-induced vasoconstriction was reduced in TRPC3 KO mice when compared to that of WT mice, but neither high K+- nor pressure-induced vasoconstriction was altered in TRPC3 KO mice. Acetylcholine-induced vasorelaxation was inhibited in TRPC3 KO mice and by the selective TRPC3 blocker pyrazole-3. Acetylcholine blocked the phenylephrine-induced increase in Ca2+ ratio and then relaxation in TRPC3 WT mice but had little effect on those outcomes in KO mice. Acetylcholine evoked a Ca2+ increase in endothelial cells, which was inhibited by pyrazole-3. Acetylcholine induced increased NO release in TRPC3 WT mice, but not in KO mice. Acetylcholine also increased the nitrate/nitrite concentration in TRPC3 WT mice, but not in KO mice. The present study directly demonstrated that the TRPC3 channel is involved in agonist-induced vasoconstriction and plays important role in NO-mediated vasorelaxation of intact mesenteric arteries.Fil: Yeon, Soo-In.     An Empirical Investigation of Smart Product Adoption

    The advance of information technologies and the Internet have been enabling the transformation of physical products into smart products by embedding information technologies into the products and thereby making them intelligent. The movement to the ā€˜Internet of Thingsā€™ is accelerating connection of the products to the net. While those changes could enhance value propositions of products, they might also cause consumer privacy concerns, which might hinder smart product adoption, because the smartness of the product mainly takes advantage of personal information about the users. This study aims to investigate consumersā€™ intention to adopt smart products. Building on previous studies on smart products and privacy literature, we propose a research model that explains factors influencing consumersā€™ intention to adopt smart products. The proposed research model is empirically tested using data from an online survey of consumers. The overall results validate the proposed research model of smart product adoption. Specifically, perceived personalization is found to positively affect consumersā€™ intention to adopt smart products, whereas information privacy risk decreases the intention. We also find that the attributes of personal information are critical antecedents of consumersā€™ risk-benefit assessment. The sensitivity of information increases information privacy risk while the congruency of information enhances perceived personalization. Based on the results, theoretical and managerial implications are discussed

    Singlet Fermionic Dark Matter with Dark ZZ

    We present a fermionic dark matter model mediated by the hidden gauge boson. We assume the QED-like hidden sector which consists of a Dirac fermion and U(1)X_X gauge symmetry, and introduce an additional scalar electroweak doublet field with the U(1)X_X charge as a mediator. The hidden U(1)X_X symmetry is spontaneously broken by the electroweak symmetry breaking and there exists a massive extra neutral gauge boson in this model which is the mediator between the hidden and visible sectors. Due to the U(1)X_X charge, the additional scalar doublet does not couple to the Standard Model fermions, which leads to the Higgs sector of type I two Higgs doublet model. The new gauge boson couples to the Standard Model fermions with couplings proportional to those of the ordinary ZZ boson but very suppressed, thus we call it the dark ZZ boson. We study the phenomenology of the dark ZZ boson and the Higgs sector, and show the hidden fermion can be the dark matter candidate.Comment: 10 pages, 3 figure
